1873年，美国马里兰州巴尔的摩市银行家、教友派教徒约翰 ·霍普金斯去世时，留下了一笔当时价值700万美元的巨额遗产。遵照他的遗嘱，其遗产分别捐赠给以他名字命名的大学和医院。1875年，其财产托管人在巴尔的摩市中心的霍华德大街购买了第一块地皮，以此作为未来的大学校园。1876年2月22日，正式创立约翰·霍普金斯大学。
